Ozempic: Scientists raise alarm over its possible impact on the brain
Ozempic has become one of the most widely used drugs in the world in recent years, mainly for weight loss and diabetes control. But while millions of people use it to reduce appetite and excess weight, scientists are discovering that its effects may go far beyond the stomach and metabolism, directly affecting the brain as well.
One of the studies that has drawn attention is led by Alison Shapiro, an assistant professor at the University of Colorado Anschutz. She was part of a research team that studied 13 adolescent girls and young women with hormonal disorders who were given GLP-1 drugs such as Ozempic.
As part of the research, the scientists conducted brain scans before and after treatment. The results were surprising: within a few months, significant changes were observed in brain neural connections, especially in the so-called “salience network,” a system linked to attention, focus, and information processing.
“We did not expect to see this result, and we still don’t really know what it means,” Shapiro said, stressing that the research is still in its early stages.
From a metabolic drug to a subject of neuroscience research
Initially, GLP-1 drugs such as Ozempic were considered a revolution in the treatment of diabetes and obesity. They mimic natural hormones in the body that help control hunger, blood sugar levels, and digestion.
But scientists are now beginning to understand that their impact may be far more complex. Tens of millions of people worldwide are using these medications, turning the phenomenon into one of the largest “unintentional experiments” in modern medicine in the field of neuroscience.
Experts are studying whether GLP-1 affects not only how people eat, but also:
addictive behaviors,
cognitive functions,
neurodegeneration,
mood and emotional pleasure.
Physical and mental health increasingly interconnected
Recent research shows a major shift in how science views the body and the brain. Researchers are increasingly moving away from the idea that physical and mental health are two separate fields.
According to experts, the hormones and receptors activated by GLP-1 drugs create a communication network that extends beyond the stomach. Their receptors are found not only in the digestive system, but also in the heart and in certain areas of the brain.
This has raised important questions:
Do these drugs directly affect the brain?
Can they change the way neural networks function?
Can they influence emotions, motivation, or pleasure?
Unusual effects reported by users
While many users report significant weight loss and reduced appetite, not all reported mental effects have been positive.
On social media and in medical reports, some users have described:

* reduced enjoyment of everyday activities,
* loss of interest in hobbies,
* decreased sexual desire.
Some have described this condition as a “flat emotion,” where pleasure and motivation appear reduced.
How might Ozempic affect the brain?
Scientists still do not have a definitive answer. One of the main reasons is that GLP-1 drugs are relatively large molecules, and it is still not fully known how much of them can cross the blood–brain barrier, the protective membrane that separates the brain from the bloodstream.
However, several theories are gaining traction:
Reducing inflammation in the brain
Researchers believe GLP-1 may help reduce inflammation that, over time, can damage neurons.
Neuroprotective effects
Other studies suggest these drugs may help brain cells function better and cope more easily with biological stress.
Gut–brain signaling
Another theory involves the vagus nerve, the main communication pathway between the digestive system and the brain. Scientists suspect that changes in appetite and digestion may indirectly affect mood, desire, and cognitive function.
Experts emphasize that research is still ongoing and there is currently insufficient evidence to draw firm conclusions about the long-term neurological effects of Ozempic and similar drugs.
However, the possibility that these medications may affect not only the body but also the brain is opening a new chapter in modern medicine and neuroscience.
